Fluorescence-Based Automated Cell Counter with PI Staining
The ADAM MC automated cell counter performs viability and cell counting measurements using the PI (Propidium Iodide) staining method for dead-cell staining combined with advanced image analysis. Based on fluorescent microscopy techniques, ADAM utilizes sensitive fluorescence dye staining, LED optics and CCD detection technologies to deliver accurate and reliable cell analysis.
To count cells, samples are mixed with Propidium Iodide (PI) stain and pipetted onto a disposable plastic chip. The chip is loaded onto a precision stage where the ADAM system automatically focuses, records images with a sensitive CCD camera, and processes the results—displaying the cell count automatically. Simple. Fast. Accurate. Reliable.
Technology & Accuracy
As Accurate as FACS Counting
ADAM analyzes 60 images equivalent to an 8.5 µL counting volume—8.5 times more accurate than 1 µL analysis. This addresses variation from low sample reads, low sampling volumes, and clumped cells. Results commonly show R² values over 0.99 and CVs under 1%.

Propidium Iodide Staining Method
ADAM-MC stains mammalian cell DNA with Propidium Iodide (PI), which does not enter cells with intact membranes or active metabolism. Only non-viable cells with damaged membranes or inactive metabolism allow PI to enter and stain the nucleus.
AccuStain Solutions:
Solution T (Total Cell Counting): Contains PI and lysis solution to disrupt all plasma membranes and stain all nuclei for total cell count.
Solution N (Non-viable Cell Counting): Contains PI and PBS. Live cells remain intact and unstained; only non-viable cells are stained.
After treatment, samples are loaded into the chip and viability is automatically calculated by ADAM-MC software after measuring both total cells and non-viable cells.
